INTRODUCTION

The RNAscope® Multiplex Fluorescent Assay is based on ACD’s patented signal amplification and background suppression technology. The assay uses a novel and proprietary method of in situ hybridization (ISH) to visualize single RNA molecules per cell in fresh frozen tissue samples mounted on slides. RNAscope® Assays do not require the RNA-free environment used for traditional ISH. This RNA ISH assay provides the same exceptional sensitivity as our singleplex assays, allowing single-molecule detection of up to three RNA targets simultaneously and is ideal for co-localization studies of any expressed gene set in nearly any tissue type. Up to 4 fluorescent labels are used including FITC (green), Cy3 (orange), Cy5 (far red)  and Cy7 (infared).  

RNAscope® Multiplex Fluorescent Reagent Kit is equipped with reagents to multiplex three probes using FITC (green), Cy3 (orange), Cy5 (far red) dyes .  If your assay requires fourplex, ACD can provide custom target probes for the 4th channel using Cy7 (infared) dye.  Additionally we have a RNAscope® Multiplex Custom Kit. 

The RNAscope® ISH assay is compatible with a variety of sample types including fresh frozen or formalin fixed parrafin embedded slides or tissue blocks, fixed frozen slides, chamber slides contains adherent cells, peripheral blood mononuclear cells and circulating tumor cells.  This assay is most ideal when used with fresh frozen tissue and chamber slides contains adherent cell.

The RNAscope® Multiplex Fluorescent Assay workflow requires:

  • RNAscope® Target Probes (Probe names are designated with C1, C2, C3)

  • RNAscope® Control Probes (Probe names are designated with C1, C2, C3)

  • RNAscope® Control Slides (optional)

  • RNAscope® Multiplex Fluorescent Reagent Kit 

Each RNAscope® Multiplex Fluorescent Reagent Kit provides enough reagents to stain ~20 tissue sections approximately 20 mm x 20 mm large. Larger tissue sections will result in fewer tests. Each kit contains three sub-kits: a Pretreatment Kit, a Detection Kit, and  Wash Buffer.
